Businesses, libraries and universities can substantially grow by using strategies to overcome cloud computing challenges identified in this study. 1. Introduction 1.1 History and Development of â€Å"Cloud† Development of â€Å"cloud† can be considered as an inevitable trend which began in 1950s as hardware time-sharing (Earnst and Young, 2012) and â€Å"Utility computing† and â€Å"Grid computing† in late 1980s (Global Technology Industry, 2011). Thus aggregating assets had been beneficial over on-premise computing since the beginning of IT industry. â€Å"Utility computing† allowed consumers to pay for computer software based on the amount they utilized as opposed to the traditional method of purchasing the entire software. â€Å"Grid computing† aggregated the processing power of several idle computers, decreased the time taken for operations of large software and provided multiple accesses to the data bases stored in remote computer labs, libraries and other offices in the universities (Educause, 2006). Moreover, Utility computing and Grid computing decreased the cost of purchasing license for software, increased convenience in monitoring the information systems in universities, aggregated the processors and decreased the time consumption on detailed-software such the 3D graphic designing. But these concepts were restricted to small scale operations in these organizations. Development of cloud enabled large scales businesses reaping benefits from resource aggregation and virtualization. It contains similar characteristics to Utility computing, Grid computing, High Speed Computers and clusters. But cloud computing essentially defers from Utility and Grid computing, due to many attributes. Example: services, deployment, associated issues and scope for development. These are described in detai l in the proceeding chapters. Furthermore, public used remotely hosted servers for connecting to the internet and obtaining web services such as Gmail, chat, e-banking and social-networking in large scale for a considerable time period in the history. But resource virtualization was not recognized as a possible venture involving vast economic benefits until the development of cloud in recent history. Cloud computing involves large scale aggregating and vitalizing of IT assets. It delivers a number of economic and operational benefits to all the consumer segments (Gong, Liu, Zhang, Chen and Gong, 2010). It has a growing potential to decrease the firms’ production costs, create virtual business firms and market business process as a service in modern economies (Earnst and Young, 2012). As opposed to Grid computing which mainly involved academic and government institutions cloud computing service providers involve commercial IT giants such as Google, Microsoft, Apple, Amazon, IB M and Oracle (Gong et. al, 2010). And also private, cooperate and contract software developers and venders also market in clouds. Consumers of cloud computing services vary from small to large scale business firms, universities federal to individuals. For over 40 years, the link between inflation and unemployment has been intensely debated upon by economists all over the world (Oliver, 1999). They hypothesis postulated by Phillips has been subjected to myriad criticisms regarding its ability to explain the inflation-unemployment relation. For instance, in the 1970s, the Phillips curve fell short of elucidating why many countries experienced stagflation - an economic condition characterised by high level of unemployment coupled with high level of inflation ("Wikipedia," 2005). With this, new theories emerged to better illustrate the observed link between inflation and unemployment levels. One of the most notable is the theory on non-accelerating inflation rate of unemployment (NAIRU) or the natural rate of unemployment. Based on this theory, the short term Phillips curve is negatively sloping showing the inverse relationship between unemployment (on the x-axis) and inflation (on the y-axis). (Samuelson & Nordhaus, 2001) On the other hand, the long run Phillips curve is vertical, wherein only the natural rate of unemployment was consistent with stable inflation rate as expectations of market players change. In this regard, no trade-off is seen between inflation and unemployment in the long run. However, this theory is critiqued due to the difficulty in determining the natural unemployment rate. (Levacic & Rebmann, 1982) Another theory, which is influential in explaining the inflation-unemployment linkage, is the one postulated by Milton Friedman. He argued that the Phillips curve is formed due to "money illusion" such that the price inflation "fooled" businesses into perceiving that there is demand surge. As such, they hire more people, thus, decreasing the unemployment rate. Friedman's theory asserted that inflation tends to precede drops in unemployment rather than follow it. (Oliver, 1999) Other economists believe that, contrary to Friedman's theory, low unemployment raises bargaining power of workers. Given this, they tend to push for higher nominal wages resulting in cost-push inflation. Employers then would raise prices to remain within targeted profit level. My Summer Vacation Essay Relaxing out in the sun in the middle of a lake that stretches for miles and miles, is probably one of the most amazing things you could ever see in life. The best part about that, I get to enjoy that every summer. Every summer my family goes to a small cabin on Lake Rainy in Canada. The most memorable time I have ever had with my family was at our family cabin in Canada. It was the summer of 2007 and also one of the hottest trips weve had at our cabin. One of my favorite things about the cabin is that it has no electricity unless we have the generator on. My brothers whined about that for the first couple days! Bubba even tried to convince Aunt Linda to turn on the generator just so he could watch a movie. With no success with my aunt he asked Uncle Tom and got his way. After the kids got the idea that there would be no more turning the generator on for little things like charging their Game Boys, things actually started to get better. With almost all technology set aside we did things unheard of in this day in age. One night we decided we were tired of being bored and I thought my little brother to play Kings Corner, they didn’t like that game very much. I decided it was time for a little easier game; it would have to be easy to catch Bubbas attention. Uno was the game we played most nights. Playing Uno at the cabin was great, the entire family was ordered to play by my Aunt Linda and since she is the boss, no one would have even thought otherwise. Don’t get me wrong we didn’t spend all of our time indoors. We went swimming very frequently, most of the time just to stay cool. It was usually too hot to go fishing, unless you had a death wish. One day when it was cool enough to fish, Kelsey caught the most beautiful Walleye I have ever seen. My little cousin Kimmy wasnt to excited about eating fish so the made her chicken every night we had fish. (This was most nights. ) The only thing I hated about Canada honestly was leaving. On the last day were at the cabin everyone is usually pretty sad to be leaving. It was always nice to get home, but as soon as we would get home we would instantly start to miss watching the sunset on a lake that stretched for miles and miles.

A very important point is where Alfieri is talking about men in the Mafia who were shot by thugs from other families, and refers to those men as justly shot by unjust men which means that those men deserved to die, yet they shouldnt, in the eyes of the American Judicial System, be killed without trial, that being one of the Amendments of The Constitution. Perhaps the most important occurrence when dealing with this category is the prefiguration about Vinnie Bolzano (which has been previously mentioned), because it serves as a prefiguration about what happens when someone breaks the Code of Conduct, with regards to immigrants. The next type of tension is about Eddie as a sympathetic character. Miller wanted the audience to feel sympathetic for Eddie and does this by creating a sense of emotional involvement with Eddie, which forms a certain sense of empathy with Eddie, and a sorrow which implants a sorrow due to Eddies fate. This type of tension is similar to climatic tension except it creates an apprehensive mood in the audiences mind. Because this is in place early on, it is easier for the reader to justify Eddies actions and then empathise with him. In Arthur Millers view, Eddie posses or exemplifies the wondrous and human fact that he too can be driven to what in the last analysis is a sacrifice of himself for his conception, however misguided or right, dignity and justice. This to me provokes real empathy because it is very difficult to sacrifice for your beliefs, and for what behaviour he displays, he deserves so much more respect that he is given. Alfieri, upon meeting Eddie, described Eddie as having eyes like tunnels which suggests that he is in a trance like state, and he has probably given way to his emotions, which is not necessarily his fault, it may be that he is emotionally unstable, and insecure, it may be that he is suffering from stress, which makes him less mentally adept, but whatever he is, he should not, in this situation, be held accountable for his actions. Early on in the play, with Eddies conversation between Catherine Beatrice, a lot of Eddies gentle and more compassionate side is exposed when Catherine wants to work, Eddie wants her to stay in school, but she protests: Eddie: You wanna go to work, heh, Madonna? Catherine: Yeah Eddie: Alright, go to work. This shows Eddies compassionate side, because he gives in to Catherines demands, even though it is against his will, he does it because it is in Catherines interest. Another moment where Miller shows Eddies consideration for Catherine is the moment where she lights the cigar, and almost burns herself: Catherine: Here! Ill light it for you! Dont worry about me Eddie, heh? Eddie: Dont burn yourself. (Just in time she blows out match) This shows Catherines naivety, and perhaps her inability to function independently without Eddie, and how much he cares for her. She soon forgets this when she encounters Rodolfo, and Eddie feels displaced by him, like he is no longer needed, and for this reason, I think his goal becomes to have Rodolfo deported. But only because Catherine completely ignores him, and feels she is independent, when clearly, she is not. The last kind of tension is emotional tension. This is the disarray that begins due to the introduction of a stranger into this family, and continues due to Rodolfos unusual behaviour, Eddies emotional insecurities, Catherines naivety and Beatrices selfishness plunge them into. Due to analysis, I have come to realise that there is a vicious emotional cycle in place that erodes the already fragile relationship these characters share. Here is a diagram and an explanation: Catherine and Rodolfo take Beatrices advice. They become closer, get more intimate. They start to ignore Eddies opinion more. Catherine starts to become less nai ve, more aware, much to Eddies disapproval. Next Catherine and Rodolfos actions make Eddie more irate. He becomes more hypnotised by rage. He is more desperate to separate Catherine and Rodolfo. He feels displaced, like Catherine is bestowing all her affection on Rodolfo. He starts to ignore Beatrice and begins to focus solely on Rodolfos expulsion. Then Beatrice is annoyed by Eddie. She feels he is not attentive enough. She feels displace by Catherine and becomes even more jealous of the attention she receives. She encourages Catherine to become more independent and intimate with Rodolfo in the hope that she, with Rodolfo, moves out leaving Eddie to pay Beatrice more attention. This continues to occur, and destabilises this very dysfunctional family, with each party becoming more bitter and resentful to the others. It changes slightly so that Beatrice almost sides with Catherine and Rodolfo, I think purely for selfish reasons, as she has the most to gain from Catherines absence. In the end, I think is no one persons fault. I think it is the fault of all three parties. Catherine, because she is to nai ve to know what she is doing, and cannot see through Beatrices advice, Eddie for being too overprotective and getting too involved, and Beatrice, for wanting Catherine gone for all the wrong reasons. With all this, I think it is all the deceit and selfish motivation that accompanies a dysfunctional family that sees the Carbone family on their knees. In conclusion, Arthur Miller uses many types of tension in his play A View From The Bridge: climatic tension, dedicated to keeping the middle unknown; tension of discovery, to keep the play fresh with plenty of plot twists, and to keep the characters personas mysterious, the Sicilian Code of Conduct to keep the play within the same era, and to put his knowledge to good use, Eddie as a sympathetic character to keep the audience engrossed and to exploit one of the most fascinating human traits, and the triangular relationship between Eddie, Catherine and Rodolfo and Beatrice to show how easily a nuclear family can be dissolved by the introduction of a stranger who is welcomed with mixed opinions, much like a specie from a different ecosystem being introduced to a new one. Interestingly, with regard to the opinions, each member of the family represents a different opinion; Eddie represents the negative extremity (by wanting Rodolfo gone), Catherine the positive extremity (wanting to marry Rodolfo), and Beatrice who is neutral (doesnt mind, just wants Catherine gone). All these kinds of tension help to portray what happens when strangers enter a close-knit family which has mixed opinions, and that when pushed hard enough, people will forgo almost anything to protect their loved ones; their beliefs, their life, even if it really is all for the sake of self justified pride and dignity. To summarise it in one sentence, it basically says, Welcome to the human condition. Evolution of the Piano Essay One of the more popular musical instruments is the piano. In Youtube alone, a popular video sharing website, type ‘piano’ in the search box and result will show 574,000 uploaded videos. From the modern grand piano that people are grown accustomed to, it is interesting to find that piano began its journey from three things: hammer, string and soundboard. Dulcimer, an instrument originating in Iran after the birth of Christ, is said to be the inspiration for the creation of the piano (Concert Piano Pitch Services 2008). The dulcimer shares the same principles as the piano, with hammers striking/ beating strings placed in a flat soundboard (2008). However, in place of mechanical hammers, players used sticks (2008). Crusaders brought it to Europe during the Middle Ages (Marcie Wallis 2008). The term ‘dulcimer’ comes from the Latin ‘dulce melos’ which means â€Å"sweet sound† (Marcie Wallis 2008). To play, a key is depressed, sending air into the pipe, thus producing a sound (Wright 47). The pipes are arranged in groups, with each group specializing in one tone (48). A quill was used to pluck the strings. The term ‘virginal’ is said to be a compliment to the Queen. Young ladies used to play the virginal. In 1400, the world was introduced to the clavichord (Concert Piano Pitch Services 2008). It is one of the oldest keyboard instruments. It works according to a simple principle: The player depresses a key which in turns strikes a string with a metal blade called tangents, thereby producing sound (Marszalek and Panagakis 2006). It is a rectangular box with keyboards on the side and strings stretched across (Kelly 1998). Once a key is depressed, the player can also change the pitch to create a vibrato (1998). A clavichord player can make it â€Å"fretted†, striking strings in a succession manner of â€Å"unfretted† to produce one note (Piano Keyboard 2003). Widely used in Europe, clavichords were popular during the Renaissance period (1998). The Renaissance (1450-1600) was a period of rebirth, curiosity and individualism (Kamien 75). In this time, the focus was on human life (75). Thus, vocal music was given importance, with instrumental music taking a backseat (78). Given its small design, the clavichord was usually played in small audience (Get Piano Lessons. com 2008). Despite its limitations, the clavichord was a popular instrument especially in German households (2008). The oldest surviving clavichord is from the year 1543 (Piano Keyboard 2008). In this time, one instrument stood out- the harpsichord. The harpsichord originated in Northern Italy in 1400 but reached its peak during the Baroque period (Wright 48). It is the largest keyboard instrument. Some harpsichords had two keyboards (Marszalek and Panagakis 2004). Like the clavichord, the harpsichord is played by depressing a key, which in turn â€Å"forces a pick to pluck a string† (48). The plucking produces a sound (103). However, its level mechanism limits the harpsichord in controlling the forte (48). When a key is depressed, the sound and volume is still the same. Hence, harpsichords that were usually used had two keyboards to make sound gradations (48). This shortcoming exemplified the way Baroque music sounded: straightforward, uniformity continuity, no abrupt changes (103). It was during the Baroque period that Johann Sebastian Bach rose as the greatest composer, and harpsichordist of his time (Kamien 125). His Bradenburg Concerto no. 5, written around 1721, gained importance for being the first piece wherein a harpsichord had a solo role (104). Usually, in a concerto grosso (the term used for small group of soloists), the instruments used were string instruments. In the Bradenburg Concerto, the harpsichord is played in the final section. Harpsichord (Get Piano Lesons. com 2008) Henry Purcell, another famous musician from the Baroque period, was chosen to be the organist of the Chapel Royal in London (UK Piano Pages 2008). By this time, the spinet had become popular. The spinet also came from the harpsichord family (Piano Keyboard 2003). The spinet was different from the harpsichord in size and its keyboard, which was placed on the long side of instrument (2003). An image of the spinet is seen below. Image 6. Bartolomeo de Francesco Cristofori, an Italian harpsichord maker, was responsible for creating piano (2008). Instead of plucking, hammers hit the strings and a level mechanism allows the player to produce soft and loud sounds, thus the term ‘pianoforte which translates to ‘soft-loud’ (Wright 48). During the Classical Period, the piano became the key keyboard of most musicians (Kamien 150). In fact, major compositions of Joseph Haydn, Wolfgang Amadeus Mozart and Ludwig van Beethoven used the piano (150). Mozart’s Piano Concerto in A Major, one of the best known concertos of all time, was completed in this period (186). Image 7. Cristofori Pianoforte (Piano Keyboard 2003) Gottfried Silbermann, a German was touted as the maker of the first successful pianoforte (Piano Keyboard 2003). In 1742, square pianos were made (2003). The first upright piano is much more similar to a grand piano, though (2008). In 1768, Johan Christian Bach gave the first ever piano recital (2008). By 1800, John Isaac Hawkins created the first true upright piano (2008). Piano’s popularity continued until from then on. When Jazz music became popular in the 1890s to 1915, ragtime piano was developed (Kamien 360). Ragtime was played by black pianists in saloons and dance halls (360). Among the famous ragtime player at that time was Scott Joplin. One of his more prominent pieces was â€Å"The Entertainer â€Å"(360). The 1960s saw the birth of the electronic keyboard (Surovec 2002). Electric organs were used followed by amplified pianos (2002). From the spinet, virginal, clavichord and harpsichord which pluck the string, the invention of piano led to hammering the strings to produce sounds. Levers are used to work the hammers and dampers. The present piano has an iron frame which attaches the metal strings. When key is depressed, the felt-tipped hammers strike the strings, causing it to vibrate and make the sound. A piano also has tuning pins for tightening the strings. It has an 88-note keyboard and two or three pedals (Kamien 25). Half a million videos of people playing the piano is more than enough evidence that the popularity of piano has not faded. In fact, it has become more popular than ever. Music is part of our everyday lives. Be it the chirping of the birds, the machinery hum or the sound of one’s sweet singing voice. The Effects of Jazz and Classical Music on Musicians Essay A thesis presented on the history of jazz as compared to classical music and the effects on musicians, beginning with the birth of jazz, and covering the twentieth century. Berliner (1994) impresses upon the idea that jazz music is more important to a musician’s development and an individual’s mental health than classical music. It is this author’s opinion that Jazz is superior over classical music because jazz music is often soft, smooth, and reflective. In addition, the world of jazz has some wonderful artists who can both play and write jazz music so extraordinary that it will make music lovers melt like ice as the stress washes away. Jazz allows people to close their eyes, relax, mediate, and dream about their loved ones. It is the best for mediation purposes because it provides people with softness, calm, soul, and fantasy. The freedom found in jazz represents the freedom inside of all Americans. Jazz itself is a success story told through its own invention. True jazz musicians play the music that they do as a necessity to themselves. Their music is their diary, and their diary is more pure than words can tell. Jazz is Americas great contribution to the arts. It is thrilling, exhilarating, and thought-provoking music that stirs emotions of all kinds. Jazz is in the mind, heart, and in the soul. Its influence extends worldwide, and touches all related forms of music. It is an enriching art form that deserves a special place in our vibrant culture. III In the world of jazz, young performers must master a repertoire of chord progressions, off-beats, and harmonies so that they can count on them as structures around which and through which they might weave more magical variations. Jazz players use chord changes as a means of giving shape to melodic improvisation. Classical players merely read lead sheets or book arrangements and use strict chord symbols in a much more static way. â€Å"Classical music is simple for its rhythmic pattern and simplistic chords while jazz’ chords are complex† Cook (p. 17). Many may argue that classical music is a more integral part of development, but a talented classical composer will in fact draw on stylistic elements of jazz to enrich the classical tradition of symphonic music. Classical composers have consistently been making use of two elements of jazz over the years which are syncopation and harmony characterized by blue notes. â€Å"Indeed, even apparently knowledgeable classical musicians often sound confused and unsure about the essence of jazz music.† Carr (p. 174). Some of the most accomplished musicians of our time have devoted themselves to a life-long study of jazz music, and few classical musicians have been able to master jazz. Jazz music ran parallel to the development of the twentieth century classical style music. Those musicians that learn both types of music are not locked into one type of discipline, and will have more musical experiences. â€Å"We are the musical melting pot achieving a vision of merging cultures that fulfilled the image that America had of its own social destiny.† Gioia (P. 395). As a result of this concept, jazz composers began to attend symphony concerts, and subsequently jazz and classical styles began to overlap to a degree; which resulted in an art form now referred to as jazz fusion. Some would argue that jazz music is too different from classical due to being unceremonious, gritty, or too improvisational. Between 1920 and 1950 jazz and classical music together made the most successful contributions to music. â€Å"One genre essentially has the ability to learn something from the other, and can create an even more captivating sound.† Ratliff (p. 23) Many could dispute over the fact that a newcomer to jazz might feel bewildered by its proliferation of styles and differing approaches to music-making. â€Å"Indeed, harmonies are more dissonant, phrases more irregular, accents sharper, and tempos are more varied, but that is what makes it so beautiful. Its casual nature is evidenced by the inoffensiveness of wrong notes being plated, by singers taking breath sometimes without relation to lyrics, and musicians being introduced right in the middle of a performance† Ratliff (p. 72). Classical music on the other hand can occasionally trigger a thorough exploration of self and mind. It may cause a person to look at their failures in life. A sudden realization of self can be extreme or painful. Not everyone enjoys classical music. In the UK, classical music is used to drive groups of youths away from places they congregate in. Jazz music is a beautiful improvisational art making itself up as it goes along just like the country that gave it birth, and with each tap of the foot, jazz musicians reaffirm their connection to the earth. Jazz waxes and wanes between tension and repose. It challenges the musician with unpredictability and then rewarding the listener with predictable rhythms. â€Å"Jazz rewards individual expression but demands selfless collaboration.† Ward (p. 1). Listening to jazz is not just a satisfying and rewarding experience; studies show that it is actually good for a musician’s health. Listening to jazz or playing jazz melodies can relieve chronic pain and migraines, reduce blood pressure, accelerate post-stroke recovery, improve memory, boost immunity and induce relaxation. 2 Jazz music helps distract people. It gives individuals a sense of control and also releases endorphins into their bodies that alleviate pain. There is a very contextual debate that classical music is superior due to the fact it can release dopamine during anticipation and experience of peak emotion to music. It conveys very well what the composer puts into it, and although classical music does activate pleasure and reward-related regions of the brain creating a high, playing highly pitched stimulating music for long periods is not healthy because it leads to cortisol and noradrenalin secretion without the concomitant fight or flight action. Long term overdose with those hormones is well documented to create many health problems, from depression to sexual dysfunction. The current market share of Jazz in America is mere 3 percent, but jazz is still regarded as a very powerful music which is in the blood and feeling of the American people more than any other style of music. It can be made the basis of serious symphonic works of lasting value in the hands of a talented composer. â€Å"Jazz music has always been and forever will be fundamental in the development of future musicians† Cook (p. 65). Glossary Bebop: the style of jazz developed by young players in the early 40s, particularly Parker, Gillespie, Kenny Clarke, Charlie Christian and Bud Powell. Small groups were favored, and simple standard tunes or just their chord progressions were used as springboards for rapid, many-noted improvisations using long, irregular, syncopated phrasing. Improv was based on chordal harmony rather than the tune. The ‘higher intervals’ of the chords (9th, 11th and 13th) were emphasized in improv and in piano chord voicings, and alterations were used more freely than before, especially the augmented 11th. The ground beat was moved from the bass drum to the ride cymbal and the string bass, and the rhythmic feel is more flowing and subtle than before. Instrumental virtuosity was stressed, while tone quality became more restrained, less obviously ‘expressive’. The style cast a very long shadow and many of today’s players 60 years later could be fairly described as bebop. Blues: (1) A form normally consisting of 12 bars, staying in one key and moving to IV at bar 5. (2) A melodic style, with typical associated harmonies, using certain ‘blues scales’, riffs and grace notes. (3) A musical genre, ancestral to jazz and part of it. (4) A feeling that is said to inform all of jazz. Boogie (boogie-woogie): a style of piano playing very popular in the thirties. Blues, with continuous repeated eighth note patterns in the left hand and exciting but often stereotyped blues riffs and figures in the right hand. Cadence: A key-establishing chord progression, generally following the circle of fifths. A turnaround is one example of a cadence. Sometimes a whole section of a tune can be an extended cadence. In understanding the harmonic structure of a tune, it’s important to see which chords are connected to which others in cadences. Free Jazz: a style of the early and middle sixties, involving ‘free’ playing and a vehement affect. It was originally associated with black cultural nationalism. Sometimes two drummers and/or two bass players were used. Some free jazz was profound, and some not very good. Some who played it later denounced it, but the style became an ingredient in future styles and still has many proponents despite its lack of general popularity. Groove: an infectious feeling of rightness in the rhythm, of being perfectly centered. This is a difficult term to define. A Medium Groove is a tempo of, say, 112, with a slinky or funky feeling. Improvisation (improv): the process of spontaneously creating fresh melodies over the continuously repeating cycle of chord changes of a tune. The improviser may depend on the contours of the original tune, or solely on the possibilities of the chords’ harmonies, or (like Ornette Coleman) on a basis of pure melody. The ‘improv’ also refers to the improvisational section of the tune, as opposed to the head. Inner voice: a melodic line, no matter how fragmentary, lying between the bass and the melody. Interlude: an additional section in a tune, especially one between one person’s solo and another’s. The Dizzy Gillespie standard A Night In Tunisia has a famous interlude. Jazz Standard: A well-known tune by a jazz musician. See Standard. Latin: (1) Afro-Cuban, Brazilian or other South American-derived. There are many special terms used in Latin music and I haven’t tried to include them here. (2) Played with equal eighth notes as opposed to swung (see swing def. 2). Also ‘straight-8†². The feel of bossa novas and sambas. Pattern: a pre-planned melodic figure, repeated at different pitch levels. Something played automatically by the fingers without much thought. Reliance on patterns is the hallmark of a weak player. Membrane: Structure And Function Chapter title: Membrane Structure and Function. The ability of the cell to discriminate in its chemical exchanges with the environment is fundamental to life, and it is the plasma membrane that makes this selectivity possible. Membrane The membranes that are found within cells (plus the plasma membrane surrounding cells) consist of phospholipids (and other lipids plus membrane proteins) arrayed by hydrophobic exclusion into two-dimensional fluids known as known as lipid bilayers Phospholipids Phospholipids are amphipathic molecules meaning that they have both a hydrophobic and a hydrophilic end Lipid bilayer Phospholipids can exist as bilayers in aqueous solutions The hydrophobic portion of the phospholipid is shielded in middle of these bilayers The hydrophilic portion is exposed on both sides to water Lipid bilayers are held together mainly by hydrophobic interactions (including hydrophobic exclusion) Fluid mosaic model The plasma membrane contains proteins, sugars, and other lipids in addition to the phospholipids The model that describes the arrangement of these substances in and about lipid bilayers is called the fluid mosaic model Basically, membrane proteins are suspended within a two-dimensional fluid that in turn is made up mostly of phospholipids Cholesterol Cholesterol, a kind of steroid, is an amphipathic lipid that is found in lipid bilayers that serves as a temperature-stability buffer At higher temperatures cholesterol serves to impede phospholipid fluidity At lower temperatures cholesterol interferes with solidification of membranes (e.g., cholesterol functions similarly, in the latter case, to the effect of unsaturated fatty acids on lipid-bilayer fluidity) Cholesterol is found particularly in animal cell membranes Membrane proteins Proteins are typically associated with cell membranes Integral membrane proteins are typically hydrophobic where they interact with the hydrophobic portion of the membrane or hydrophilic where they interact with the hydrophilic portion of the membrane and overlying Functions of membrane proteins Functions of membrane proteins include: Transport of substances across membranes Enzymatic activity cell communication Cell-to-cell joining Attachment to the cytoskeleton and extracellular matrix Selective permeability Lipid bilayers display selective permeability In general, intact lipid bilayers are permeable to: Hydrophobic molecules (including many gasses) Small, not-ionized molecules   Simultaneously, lipid bilyaers are NOT permeable to: Larger, polar molecules (e.g., sugars) Ions, regardless of size Thus, lipid bilayers are selectively permeable barriers that allow the entry of small or hydrophobic molecules while blocking the entry of larger polar or even small charged substances Transport across membranes Movement across membranes is important, for instance as a means of removing wastes from a cell or bringing food into a cell Categories of substance transport across membranes include: Passive transport Facilitated diffusion Active transport (including cotransport) Endocytosis, phagocytosis, and exocytosis, also considered below, technically are not mechanisms of movement of substances across lipid bilayers (though these do represent movements of substances into and out of cells; to be movement across the euakaryotic cell membrane, a substance must actually pass through an endomembrane lipid bilayer) Note that in considering transport across membranes we will once again confront the concept of movement away from or towards equilibrium, i.e., endergonic and exergonic processes There are three basic types of movement across membranes: simple diffusion, passive transport, and active transport: Simple diffusion Simple diffusion is the movement of substances across lipid bilayers without the aid of membrane proteins This image (below) shows how substances move through membranes, regardless of net direction and concentration gradients: This image (below) shows how substances net move through membranes in the direction of their concentrations gradients (i.e., with their concentration gradients)-note that regardless of how net movement is accomplished, all simple diffusion across membranes occurs in the manner illustrated above, i.e., it is a process that is driven by the random movement of molecules: This figure (below) indicates the kinds of molecules that are capable of moving across membranes via simple diffusion: Passive transport Passive transport is the term used to describe the diffusion (as well as what is termed facilitated diffusion, below) of substances across lipid bilayers Passive transport is a consequence of movement through the lipid bilayer (whether by diffusion through the membrane or with movement across facilitated by an integral membrane protein) a concentration gradient thereby contrasting with active transport Down the concentration gradient Diffusion is a random process that tends to result in the net movement of substances from areas of high concentration to areas of low concentration This includes movement from one side of a permeable lipid bilayer to the other from the higher concentration side to the lower concentration side (i.e., passive transport) Movement from high to low concentration areas is described as going down its concentration gradient. The direction of movement of substances across lipid bilayers by passive transport is controlled by concentration gradients Osmosis Movement of water across selectively permeable membranes down the water concentration gradient is called osmosis Note that this is movement toward equilibrium (exergonic process) Tonicity (isotonic, hypertonic, hypotonic) Picture a membrane separating two solutions, one side with a higher solute concentration than the other The side with the higher solute concentration is said to be hypertonic The side with the lower solute concentration is said to be hypotonic (I keep track of the difference by recalling that a hypodermic syringe is so named because the tip of the needle is placed beneath the dermis, i.e., under the skin; a hypotonic solution has a solute concentration that is beneath, i.e., lower than that of the reference solution) If both sides have the same solute concentration, they are said to be isotonic Animal cells and tonicity Normally animal cells are bathed in an isotonic solution Placement of an animal cell in a hypertonic solution causes the cell to shrink (i.e., water is lost from the cell by osmosis) Placement of an animal cell in a hypotonic solution causes it to take on water then burst (lyse, i.e., die) (water is gained by the cell, lost from the environment bathing the cell, both by osmosis) Turgidity Normally a plant cell exists in a hypotonic environment The hypotonicity causes the plant cytoplasm to expand However the plant cell does not lyse and this is due to the presence of its cell wall This conditions is known as turgidity (i.e., the pressing of the plant plasma membrane up against its cell wall) Plant cells prefer to display turgidity Plasmolysis A plant or bacterial cell placed in a hypertonic environment will show a shrinkage of its cytoplasm This shrinkage is called plasmolysis At the very least plasmolysis will inhibit growth Often plasmolysis will lead to cell death This is the principle upon which foods are preserved in highly osmotic solutions (e.g., salt or sugar); such solutions impede most microbial growth Flaccidity Plant cells bathed in isotonic solutions will fail to display turgidity Instead they display flaccidity At a whole-organismal level, flaccidity is otherwise known as wilting Transport proteins Substances (e.g., sugars) that are not permeable through lipid bilayers may still cross via membrane-spanning transport proteins Facilitated diffusion Facilitated diffusion is the movement of a substance across a membrane via the employment of a transport protein, where net movement can only occur with the concentration gradient, is called facilitated diffusion The key thing to keep in mind is that facilitated diffusion, in contrast to other mechanisms of transport-protein-mediated membrane crossing, does not require any input of energy beyond that necessary to place the protein in the membrane in the first place (i.e., facilitated diffusion is an exergonic process) Passive versus active transport Two general categories of transport across membranes exist: Those that dont require an input of energy (passive transport, simple diffusion, facilitated diffusion) Those that do require an input of energy (active transport) Passive Transport Active Transport Concentration gradient With (Down) Against (Up) Without Integral Protein Yes (Simple Diffusion) No With Integral Protein Yes   Yes Examples Small or Hydrophobic Substances, Osmosis(by simple diffusion) or Not-Small or Charged Substances (by facilitated diffusion) Cotransport, Proton Pump, Sodium-Potassium Pump Active transport Active transport is the movement of substances across membranes against their concentration gradients Moving things against their concentration gradients requires an expenditure of energy (i.e., it is an endergonic process) This energy can be in the form of ATP (e.g., sodium-potassium pump) This energy can also be in the form of electrochemical gradients (i.e., cotransport) Note that the movement of substances by active transport is in a direction that is away from equilibrium Sodium-potassium pump One means by which cells actively transport substances across membranes is via the sodium-potassium pump The sodium-potassium pump is important especially in animal cells, and is the means by which the sodium-potassium electrochemical gradient is established by these cells Proton pump The sodium-potassium pump is the means by which animal cells generate membrane potentials In bacteria, plants, and fungi, proton pumps play the same role The proton pump is simply ATP-driven active transport in which the substance pumped across the membrane is a hydrogen ion Cotransport Much of the active transport accomplished by a cell isnt directly powered by ATP Instead, much active transport is powered by membrane potentials (i.e., electrochemical gradients) Such electrochemical-gradient-driven active transport is called cotransport In cotransport, one substance, such as a sugar, is driven up its concentration gradient while a second substance, e.g., sodium ions or protons, are allowed to fall down their electrochemical gradient; the energy gained from the latter is employed to power the former (i.e., energy coupling) Endocytosis Endocytosis is a general category of mechanisms that move substances from outside of the cell to inside of the cell, but neither across a membrane (technically) nor into the cytoplasm (again, technically speaking) Instead, substances are moved from outside of the cell and into the lumens of endomembrane system members To enter the cytoplasm an endocytosed substance must still be moved across the membrane of the endomembrane system, e.g., following their digestion (typically hydrolysis) to smaller molecules Examples include: phagocytosis, pinocytosis, and receptor-mediated endocytosis Phagocytosis Phagocytosis is the engulfing of extracellular particles is achieved by wrapping pseudopodia around the particles, thus internalizing the particles into vacuoles Amoebas employ phagocytosis to eat Most protozoa obtain their food by engulfing, i.e., via some form of endocytosis The advantage of endocytosis as a mechanism of food gathering has to do with minimizing the volume within which digestive enzymes must work in order to digest food, i.e., the engulfed food particle Cells in our own bodies, called phagocytes and macrophages employ phagocytosis to engulf (and then destroy) debris floating around our bodies as well as to engulf and destroy invading bacteria Pinocytosis Pinocytosis is the engulfing of liquid surrounding a cell This is how developing ova obtain nutrients from their surrounding nurse cells (ova are very large cells so have surface-to-volume problems-pinocytosis solves the problem of nutrient acquisition by allowing nutrients to be obtained across many internal membranes rather than being limited to crossing the plasma membrane) Receptor-mediated endocytosis Receptor-mediated endocytosis involves the binding of extracellular substances to membrane-associated receptors, which in turn induces the formation of a vesicles Exocytosis Exocytosis is more or less the mechanistic opposite of endocytosis
